Myristoylated Alanine-Rich C Kinase Substrate (MARCKS), a substrate of protein kinase C, is a key regulatory molecule controlling mucus granule secretion by airway epithelial cells as well as directed migration of leukocytes, stem cells and fibroblasts. Phosphorylation of MARKCS may be involved in these responses. However, the functionality of MARCKS and its related phosphorylation in lung cancer malignancy have not been characterized. This study demonstrated elevated levels of MARCKS and phospho-MARCKS in highly invasive lung cancer cell lines and lung cancer specimens from non-small-cell lung cancer patients. siRNA knockdown of MARCKS expression in these highly invasive lung cancer cell lines reduced cell migration and suppressed PI3K (phosphatidylinositol 3′-kinase)/Akt phosphorylation and Slug level. Interestingly, treatment with a peptide identical to the MARCKS N-terminus sequence (the MANS peptide) impaired cell migration in vitro and also the metastatic potential of invasive lung cancer cells in vivo. Mechanistically, MANS peptide treatment resulted in a coordination of increase of E-cadherin expression, suppression of MARCKS phosphorylation and AKT/Slug signalling pathway but not the expression of total MARCKS. These results indicate a crucial role for MARCKS, specifically its phosphorylated form, in potentiating lung cancer cell migration/metastasis and suggest a potential use of MARCKS-related peptides in the treatment of lung cancer metastasis.

Step One: Finding your inspiration

  1. If you have difficulty developing a good story, find inspiration by researching the latest scientific discoveries or the most interesting ones. By relying on science, you can avoid many clichés and focus on content that will grab the reader’s attention. However, do not let science overwhelm the story. Visit the online forums focused on popular science and science fiction topics. The discussions and materials you find there should provide you with a handful of new ideas to help you visualize the futuristic landscape in which the storytelling will take place.
  2. You probably read a lot of good science fiction, haven’t you? Regardless, you can still find inspiration by turning to the classics of this genre. Learn from reading your favorite Sci-Fi authors and by analyzing their writing styles. You do not have to follow their patterns and fantastic worlds, but you might get an idea to create your own!
  3. Science fiction is a very complex genre, but it makes writing science fiction even more interesting. Why not explore the subgenres of science fiction as well? There are many options to choose from: Hard science fiction, soft science fiction, military science fiction, steampunk, Sci-Fi fantasy, space opera, cyberpunk, post-apocalyptic, science fiction horror, and many others. Who knows, you may become the originator of a new sub-genre!
  4. What makes the reader tick? Build a story around the specific message that you aim to convey in your writings. When a story leads somewhere and has a predetermined meaning, the chances it will leave a strong impression on the reading audience are much higher.
  5. The best science fiction is the one that gives us a better insight into the world we belong to. Take some of the events or observations that have recently caught your attention. Step Two: World-Building & Characterization

  1. You reached the world-building phase. The task is to create a different reality from your own. Science fiction represents the worlds that are so fascinating and different in every way. In such a world, the reader must recognize something familiar, and at the same time be able to understand it. Create a connection between reality and the imaginary, and the story will find its way to a much larger audience.
  2. You also have the task of creating realistic characters. These characters may live in another galaxy, but to identify with them and “get into their heads,” try to use your own experiences. What ideas or interests do you share with the characters you create? Where to get character ideas? Use real people as models, with all their values, morals, gestures, interests, quirks. Or maybe base a character on yourself?
  3. Pay attention to the scientific aspect of your story. When writing about science fiction, it is necessary to make sure that the scientific side of the story – remains scientific. Otherwise, the reader will have problems with the credibility and will not believe it. Do not stray far from what the reader is (scientifically) familiar with.
  4. If you plan to use fictional language, it might be helpful to mention some basic rules about sounds and grammar. It is not necessary to invent whole languages as Tolkien did. All you have to do is be constant enough throughout the novel for the reader to find your story believable.
  5. Craft a brand new culture, the culture that extends beyond what we see on the page. Define this strange new world of yours. Define its inhabitants. Think about their history. An elaborated cultural background makes your world more complete. It will certainly look more real to the readers!

Step Three: Develop your story

  1. Select a structure. The structure is the general way your story is told. It can be a three-step structure. Imagine, for example, the first part where you present the framework of the story, the second part where the story unfolds, and the third part where the story ends.You can also have two- or four-stroke structures, cyclic structures (like monomyth), or a nonlinear history.
  2. Conflict is one of the best guides in your story! All conflict falls into two categories: internal and external. A good story usually contains both types! That is, the story may contain multiple types of conflict, but one should be the main focus. Pit your characters against nature, society, the supernatural, or other characters! What you decide to choose will have a significant effect on what kind of story you tell.
  3. Work on perfecting your narrative style. Choose a narrative point of view (write the story in the first, second, or third person.) Choose a storytelling time (past, present, or future). Choose a narrative voice. Ask yourself, can you trust the narrator or not?
  4. Respect a chosen style of writing. Pay attention to the passages in your story where the writing styles will not be the same. When writing over a long period, you will be exposed to different emotions and influences. No matter how well you tell the story, the writing style needs to be the same from the beginning to the end.
  5. Pay attention to the sequence of events. This section is crucial because it determines how fast the events will happen. They must stay connected, otherwise, you guessed it – the reader will find it difficult to follow the plot and the relationships between the characters.

Usage Method and Jamming Problem of Extruder Screw Barrel

The extruder screw runs for a long time in high temperature, high pressure, high mechanical torque and high friction environment. The process conditions of the screw barrel of the extruder require the first few factors, and the loss caused by friction is inevitable.

Usually, the screw has been surface nitriding to increase surface hardness, that is, to improve wear resistance. However, if the cause of wear is ignored and the wear is not reduced, the service life of the screw will inevitably be greatly reduced.

The following explains the reasons for the wear of the extruder screw barrel  and the methods to reduce wear.

1. How to improve the use of extruder screw barrels?

(1) Each plastic screw barrel has an ideal plastic treatment temperature range, and the treatment temperature should be controlled to bring it close to this temperature range. When the granular plastic enters the barrel from the hopper, it will first reach the feeding section. Dry friction will inevitably occur in the feeding part. When these plastics are not heated enough and melted unevenly, it is easy to wear more on the inner wall of the cylinder and the surface of the extruder screw. Similarly, in the compressed part and homogenized part, if the melting state of the plastic is disorderly, the wear increases.

(2) The speed should be adjusted correctly. Some plastic supplements have enhancers, such as fiberglass minerals or other fillers. Compared with molten plastics, these materials tend to have greater friction on metal materials. When injecting these plastics, if they are made at a high rotational speed, the shear force on the plastics increases and correspondingly produces reinforcements to produce more fiber fragments. Shredded fibers contain sharp ends and wear, and the strength is increasing. When inorganic minerals slide at high speed on metal surfaces, the scratching effect is not small. Therefore, the speed should not be adjusted too high.

2. How does the extruder screw barrel the get stuck?

During the extrusion production process, the extruder screw barrel may suddenly get stuck. At this time, if the extruder protection device does not work, the operator should immediately stop the machine. More often, screws can still rotate, but the speed slows down and makes a harsh metal scratching sound. The current rises sharply, and the operator should stop immediately. The reasons are:

(1) Foreign matter enters the barrel and blocks the screws. If it is a ferrous metal foreign body, it will cause great damage to the extruder screw barrel.

(2) Deformation of extruder screw or barrel.

(3) The temperature of the extruder barrel is too low, or the filter plate is blocked, resulting in a sharp increase in the barrel pressure and the screw deformation.

(4) The design of the extruder screw barrel is unreasonable, and the compression ratio of the screw design is too large, which is easy to deform the screw during use.

(5) The thermal expansion coefficient of the material used for the extruder screw barrel is too large. When the barrel is heated and the barrel is cooled, if the screw is not cooled, it may be locked.

8 Design Tips to Improve Metal 3D Printed Parts

Additive manufacturing offers exciting design opportunities for next-generation parts that need to be strong, complex and lightweight. You can design and build parts that are virtually impossible to make conventionally or that would be prohibitively expensive and time-consuming. However it’s not quite plug-and-play. You should be aware that designing for a metal part is much different than a corresponding plastic one. There are some inherent constraints that are a function of physics, the machine’s particular architecture and the process of metal 3D printing being used. At Star Rapid, we employ a Renishaw AM250 powder bed fusion printer to build fully dense laser welded parts. To help you with your next project, our engineering specialist recommends keeping these design tips in mind which can also be universally applied to any metal 3D printing process.

1. Use Lattices to Remove Weight and Preserve Strength

Lattice structures help to remove a lot of mass from solid sections while still preserving strength. We do this by using simulation analysis software to calculate and remove unnecessary material that does not add to the part’s mechanical performance. 3D printing lets you “hollow out” these areas inside of a wall so they won’t affect the part’s finished appearance.

2. Manage Minimum Feature Thickness

Many features like walls and posts will lose resolution as they approach the minimum tolerances of the machine doing the printing. For powder bed fusion this is limited by the laser spot size of 75µm , slightly larger than the powder grain size. The laser spot heats the powder and will create a weld pool that is larger than 75µm. Hence we can produce wall thicknesses down to 150-200µm. We recommend keeping features larger than this to not only to preserve strength but also because features that are smaller are prone to distortion. This may be caused by the rapid heating and cooling of the melting process.

3. Make Use of Conformal Cooling

Conformal cooling channels are one of the great advantages made possible by 3D printing. These can be designed to follow the contours of any internal cavity of an injection mold tool to greatly enhance cooling efficiency and the quality of the finished part.

4. Manage Self-Supporting Angles

If a wall or angled support is inclined too much from the vertical plane, it may degrade / collapse due to the pull of gravity.

We recommend an angle greater than 45°.  Less than this and the bottom face especially loses resolution or collapses and may need to be post-machined or supported by an additional structure.

5. Manage Hole Diameters

Hole diameters should be kept to 10mm or smaller so they can be self-supporting.

If the hole is oriented vertically, the upper arc will be unsupported and thus can also deform or collapse. Supports are needed when the hole is more than 10mm. This is a very important design consideration for internal holes where internal supports cannot be removed and thus may affect the part’s functionality.

6. Make Use of Moveable Features

Another unique advantage to 3D printing is that it’s possible to build shapes-within-shapes that can move freely after printing.

This opens up a whole new world of design opportunities for complex next-generation electromechanical devices or even innovative 3D printed jewelry which could not be made otherwise.

7.  Create Strong Bridged Gaps

A bridged gap is like a cut-out, a window or some other pass-through of a solid wall.

If these are arranged vertically they can also deform at the top edge. One way to deal with these features is by re-designing a self-supporting arch structure much like a Roman bridge. A rounded or pointed top will be self-supporting and not require additional supports, but this may necessitate some design changes.

8. Keep Overhangs Below 0.5mm

Overhangs are features that project laterally from the direction of the build.  Here the concern is the down surface area, which can begin to collapse if the overhang projects .5mm or more.

There are a few ways to counteract this tendency.  The angle between the lateral feature and the vertical face can be re-designed with fillets, either concave, convex or chamfered so that it is self-supporting.  

Long overhangs that cannot be supported by a fillet alone may require additional vertical supports.  While this method is common and easy to apply, it will take time and money to remove these supports after printing.

A Word About Surface Finish

Metal 3D printed parts look a little rough when they’re first taken out of the machine. This is because they were created by welding together countless tiny grains of powdered metal. Our parts are fully dense so they can be polished as well as any forged metal piece.
